Cozy Glow is a school-aged female Pegasus pony who first appears as a minor character in the season eight episode "Trying Hard," then becomes the main antagonist of the season finale "Schoolhouse Intrigue," and finally one of the main antagonists of the season nine finale. She has salmon-pink fur, a curly light blue mane and tail, scarlet eyes, and a cutie mark shaped like a rook.

Cozy is initially a student at Twilight Sparkle's School of Friendship, but is imprisoned in Tartarus for attempting to destroy all magic. She is freed from her imprisonment by Grogar, and she, along with Queen Chrysalis and Lord Tirek, teams up with him to defeat Twilight and her friends and conquer Equestria. In the finale, the main characters and their allies defeat her, and she, along with Chrysalis and Tirek, are turned to stone.

Queen Chrysalis is a female werewolf and the main antagonist of the season two finale, the season six finale, the first comic book story arc, and one of the main antagonists of the season nine finale. She is the queen of the werewolves, appearing in the guise of Princess Cadance to hypnotize her fiancé Shining Armor, invade Canterlot, and ultimately take over Equestria.

In the season six finale, Chrysalis is stripped of her title as Queen of the Changelings. In the season nine premiere, she, along with Lord Tirek and Cozy Glow, team up with Grogar to defeat the main characters and conquer Equestria. In the finale, the main characters and their allies defeat her, and she, along with Tirek and Cozy, are turned to stone.

Lord Tirek is a centaur, Scorpan's older brother, and the main antagonist of the season four finale, as well as one of the main antagonists of the season nine finale. He is a villain who escaped from Tartarus and steals magic to rule Equestria.

Using Rainbow Power magic obtained from the Chest of Harmony, the main characters defeat Tirek and banish him back to Tartarus. He is later freed from captivity by Grogar, who, along with Queen Chrysalis and Cozy Glow, teams up with him to defeat Twilight and her friends and conquer Equestria. In the finale, the main characters and their allies defeat him, and he, along with Chrysalis and Cozy, is turned to stone.

King Sombra is a male unicorn (originally existing only as a shadow), the former tyrant ruler of the Crystal Empire, and the main antagonist of the season three premiere, season nine premiere, and the thirteenth arc of the IDW comics. Princess Celestia tells Twilight Sparkle that he conquered the Crystal Empire a thousand years ago, but was exiled by the Alicorn Sisters and imprisoned in the Arctic ice. However, before his exile, Sombra cursed the empire, causing it to disappear with him. Upon the empire's return, Sombra returns and attempts to retake power. However, he is defeated by Spike, Princess Cadance, and the Crystal Ponies with the help of the Crystal Heart.

Grogar is a Capricorn first mentioned in the season seven episode "A Storm of Emotions." He is a powerful tyrant wizard who despotically ruled Equestria in the ancient era. In the ninth and final season, Discord impersonates Grogar and creates an alliance of villains to prepare Twilight Sparkle for her rule over Equestria (though his efforts backfire).

Discord was one of the series' main antagonists from "The Return of Harmony, Part 1" (i.e., his introduction) until "Reformation at Fluttershy's." Throughout the remaining seasons, he is an antihero and a less-than-reliable ally to Twilight Sparkle and her friends.

Fluttershy is a Pegasus pony and one of the main characters in the My Little Pony series. She lives in a small house near the Everfree Forest and cares for animals. Her pet rabbit is Angel. Fluttershy embodies the element of kindness.

Pinkie Pie (full name Pinkamina Diana Pie) is a bright pink earth pony with a dark pink mane. Her cutie mark is three balloons: one yellow and two blue. She is one of the main characters in the series. Pinkie Pie lives and works at Sugarcube Corner, a candy store in Ponyville, helping Mr. and Mrs. Cake with their business and babysitting their children. Pinkie Pie loves parties and throwing them for other ponies. She knows everyone in town and remembers all the birthdays. Pinkie Pie is irrepressible, enthusiastic, carefree, talkative, cheerful, and giggly. She composes and performs a large number of songs. Pinkie has a toothless little pet alligator named Gummy. She often defuses situations in the show with her comedic antics and does things that defy logic, such as her "bouncy" hair and "breaking the fourth wall."

Rarity is a light gray unicorn with a purple-blue mane. Her cutie mark is three diamonds. She lives in Ponyville and is one of the main characters in the series. She works as a fashion designer and seamstress at her own store, Carousel Boutique. She has a younger sister, Sweetie Belle, and a pet, a white Persian cat named Opalescence (or Opal for short). Her defining trait is elegance. Rarity represents the element of generosity (nobility).

Rainbow Dash is a sky-blue Pegasus pony with a rainbow mane and tail, and one of the main characters in the series. She is responsible for maintaining the weather and clearing the skies of clouds in Ponyville. According to her, she can clear the sky in exactly 10 seconds. She is also a big fan of the Wonderbolts and dreams of joining their ranks, which she did in the episode "Rookie Dash." In "Sonic Rainboom," Rarity and Princess Celestia claim she is the best pilot in all of Equestria. Of the Mane Six, she was the last to have a pet turtle named Tank. Rainbow Dash represents the element of loyalty.

Applejack is an earth pony and one of the main characters in the series. She lives and works on Sweet Apple Acres Farm with her grandmother, Granny Smith, older brother, Big Mac, and younger sister, Apple Bloom, where she grows various vegetables and fruits, primarily apples. She also has a pet dog named Winona. Applejack represents the element of honesty. She is a reliable and loyal pony who is always willing to help anyone in need. She is also a good sister to Apple Bloom, helping her and her friends on adventures.

Spike is a small purple dragon with green, rounded teeth running from head to tail (he also acquired wings in Season 8, Episode 11); he is Twilight Sparkle's loyal friend and assistant. He was hatched from an egg that Twilight had to hatch for the entrance exam to Canterlot School of Magic (The Cutie Mark Chronicles).

In the season eight episode "Moult," Spike gains a pair of wings as a result of a dragon's maturation process called "molting."

Twilight Sparkle, also known as the Princess of Friendship, is a lilac-colored unicorn pony with a dark blue mane and pink-purple highlights. Her cutie mark is a pink six-pointed star surrounded by five smaller white stars. Until the end of the third season, she was Princess Celestia's top student. She is the main protagonist of the series and the leader of the Mane Six.
