Approximately one thousand years before the events of the series, Marceline was born to a demon lord named Hunson Abadeer and a human woman named Elise. Abadeer is the ruler of the Netherworld. At some unknown point before she turned 5, her father separated from her and her mother. Marceline and her mother survived the events of the Mushroom War, a mysterious and devastating war that ravaged the land a thousand years before the events of the series. One of the earliest childhood memories depicted of Marceline is of her living in a trailer with her human mother, who tells her the story of how she met her father. Marceline's mother suffered from an illness, however, she tried to hide it from Marceline so as not to worry her. She asked her to draw a map of the land to reach a secret "clubhouse" where there was supposedly a party. However, as Elise's health worsened further, she told Marceline to go to the hideout alone, where she was left alone, believing that her mother had abandoned her because of her demonic powers. In the hideout there were many skeletons of other humans, and her fate was probably the same illness that Elise suffered from. During this time, Simon Petrikov found Marceline in the ruins of a city and gave her a plush toy named Hambo to cheer her up; due to the events of this episode, she is apparently 7 years old, as stated by Simon. The two developed an affectionate relationship, although the crown ruined Simon's mental health, forcing him to leave Marceline alone.

During this period of her life, Simon presumably somehow managed to reunite Marceline with her father (possibly summoning him traditionally), only to be disgusted by his cold and ruthless ways. Although she tried to tolerate his evil and selfish pranks, she eventually cut all ties with Abadeer after discovering him eating the french fries she had bought herself. In the following years, Marceline wandered through the Land of Ooo and eventually befriended a tribe of humans. These humans wore animal hats, presumably to hide the fact that they were human. At first, understandably, they were afraid of Marceline; however, after following them for some time, a member of the group, Tom Two Breads, sat down and played the guitar for the few remaining members of his group. Marceline heard this and sat down behind a bush to play the same song Tom was playing. Because of this, they saw Marceline in a brighter light and realized she was only trying to keep them safe. She became their guardian and ally, helping them kill vampires who had taken over much of the land. Marceline soon discovered that by killing a vampire, she could acquire their unique abilities thanks to her demonic power to absorb souls. The human tribe was attacked by the Vampire King while they were repairing a boat, and Marceline defended them. However, when she herself tried to fight the Vampire King, he immobilized her and bit her. Although she eventually managed to kill him, she became a vampire in the process and became the last vampire in Ooo. At some point she adopted a red poodle, from which she sucked the red, but she managed to save it thanks to the Enchiridion, and it turned white and zombie-like.

At some point, Marceline met the Sweet Princess, the ruler of the Sweet Kingdom. Marceline gave her a rock t-shirt during a musical performance, and they became romantically involved. The two often skipped official royal meetings and explored the Sweet Kingdom's elaborate rock candy mining complex, though as time went on and the increasing stress of ruling the Sweet Kingdom became more demanding, Bonnibel began to push Marceline away. This culminated when Marceline accompanied Bonnie to the Crystal Kingdom, who had asked for her help in defeating a dragon attacking her kingdom. However, Marceline didn't take the whole matter seriously and annoyed Bonnibel into paying attention to her, causing her to lash out and tell her to go home. Hurt, Marceline sang a song of anger about how she didn't care about her or her "stupid Sweet Kingdom." Marceline's song managed to defeat the dragon, but this marked the end of her relationship with Bonnie. The animosity between them would last for many years.

Later, she began a romantic relationship with a wizard named Ash. How they met is still unknown. The two would move into the Tree House, but their relationship would end after Ash sold Hambo, his most prized possession, to Maja, the Sky Witch.

Marceline eventually makes contact with Finn and Jake when she forces them to leave their home. However, she returns it after finding the cave where they found a better place for her. After this, Marceline tricks Finn into becoming her accomplice. However, Finn soon discovers that Marceline isn't evil, but rather enjoys pranks and jokes. Shortly after, the two develop a friendship.

When Marceline invites Finn to help her record a song about her father eating her french fries, Finn decides to call Abadeer to Ooo in hopes of a reconciliation. While Abadeer ravages the land, Finn discovers animosity between Marceline and her father. Finn manages to distract Abadeer by playing a recording of Marceline's "French Fry Song," leading to a brief moment of reconciliation between the two before Finn sends him back to the Nightosphere.

Marceline's ex-boyfriend, Ash, attempts to rekindle his old relationship with Marceline through deception. Disguised as Marceline's "spirit animal," he tricks Finn and Jake into believing that Marceline has fallen asleep due to a self-inflicted sleep spell. Following Ash's advice, the two enter Marceline's mind to retrieve the "memory core" in an attempt to wake her. Along the way, they encounter various memories of her childhood after the Mushroom War. However, the duo soon learns that it was an elaborate trick by Ash; Finn and Jake actually erased Marceline's memory of their breakup. In the end, Finn is able to convince Marceline, and he quickly attacks her abusive ex-boyfriend.

Marceline's father finally returns and gives her an amulet that turns her into the demonic and evil supreme lord of the Nightosphere. After a terrible experience, Finn saves Marceline from the amulet. Marceline admits that all she wants is for her father to understand her and know who she is as a person.

Some time later, Marceline seeks the help of the Sweet Princess, and the two are able to locate Maja, the titular Sky Witch. It is revealed that Ash sold Hambo to Maja the Sky Witch. Only when Bonnie changes her beloved rock shirt, which Marceline gave her, can Hambo reunite with Marceline.

After being contacted by Simon Petrikov, the original Ice King who resurfaced after losing his powers, Marceline sacrifices Hambo so she can open a time portal for Simon to reconcile with his fiancée Betty. Although the two reunite, due to the whirlwind of events, Marceline and Simon can't fully catch up before Simon reverts back to the Ice King. That said, the two manage to reunite more completely and, as such, manage to reaffirm how much they care for each other. Tired of being a vampire and wanting to "grow up," Marceline asked [him] to extract her vampire essence, in an attempt to make her mortal once more. Unfortunately, this leads to the resurgence of five of Marceline's most powerful vampire enemies: The Madman, The Empress, The Hierophant, The Moon, and the Vampire King himself. The reappearance of these vampires forces Marceline, Bonnibel, Finn, Jake, and Peppermint to stake them one by one. After finally defeating the Vampire King, his essence is eliminated; however, Peppermint accidentally spills it, causing an explosion that releases his essence. Marceline wanted to give up during this time, but the Ice King sat down with her and essentially called her a coward. This managed to motivate her to try, and she defeated the Vampire King with the help of the Sweet Princess, Finn, and Jake, but was bitten once more in the process. While turning into a vampire again, she had accepted who she was and was happy to be immortal, as she could hang out with the Sweet Princess forever.

Marceline is temporarily transformed by elemental magic into "Marshmallowine," a sentient marshmallow creature. In her candy form, Marceline is sociable and playful, but somewhat naive about the nature of the new world around her; for example, when an enraged Flame Princess attacks her, Marceline mistakes the assault. Marceline inevitably reverts to her usual state. Marceline then details her memory of these events through highly allegorical language to BMO.

Marceline's father returns to Ooo and attends one of her concerts, cheering her on too loudly in the crowd, although this was well-intentioned. While it initially embarrasses his daughter and ultimately causes a ghost fight to break out, at the end of the episode, Marceline is moved because her father was willing to support her.

She's described by Finn as a spoiled girl who loves karaoke. She seems to have a strained relationship with her biological father. Although not a villain, she can sometimes have violent and evil outbursts, as shown in "He Came from the Nightosphere," when she almost decapitates Finn near the end of the episode in an act of recklessness. Objectively, she is one of the most profound and sentimental characters. This can be linked to having been abandoned and having lived through war. She was very emotionally attached to her teddy bear, Hambo, which was given to her by the Ice King (formerly Simon Petrikov). After her ex-boyfriend Ash sold it to a witch, she became enraged and broke up with him. Later, in the episode "The Sky Witch," she recovers Hambo thanks to a trade that the Sweet Princess made with Maja.
