Donatello is a spiny softshell mutant turtle with jade green skin, a light brown plastron, and a brown and green shell with dark brown spots. The vertebrae of its spine protrude from beneath its shell and it has purple geometric markings on its shoulders and thighs. It also has a large head, yellow sclera and teeth, thick black eyebrows, three fingers on each hand, and two toes on each foot. He wears a purple bandana that covers the top of his head with short rectangular bandana tails and a pair of asymmetrical silver glasses with one red (right) and one blue (left) lens. He wears purple fingerless gloves and fingerless shoes, a silver tech glove on his left wrist with a blue touch screen, purple knee and elbow pads, a purple belt with matching pouches, and the Turtle emblem placed in the center. He also wears a purple Battle Shell that fits over his actual shell.

Donnie began life as a juvenile turtle in Baron Draxum's laboratory. Wishing to create a mutant army with formidable combat capabilities, Draxum used Lou Jitsu's DNA combined with ooze to turn Donnie from an ordinary turtle into a mutant. Later, he and his brothers were stolen from Draxum's laboratory by Lou Jitsu shortly after his mutation and Donnie was raised in the sewers of New York City alongside his brothers by Splinter. Eventually, Donnie and her siblings met a human girl named April and forged a strong bond with her over the years.
