Jacob's great-grandfather, Ephraim Black, was the last chief of the Quileute tribe, making Jacob the genetic alpha. However, because Jacob initially refused to phase or lead early on, Sam Uley stepped in to become the alpha of the main pack, with Jacob acting under him.

imprinting is an involuntary, instinctual process unique to Quileute shape-shifters where a wolf forms an instant, unbreakable, lifelong bond with a specific person.
It happens at first glance. The wolf has no choice in who they imprint on, regardless of age, preference, or existing relationships.
The wolf's world instantly revolves around the imprint. They will be whatever the person needs — a protector, best friend, brother, or romantic partner.
It is absolute law among the wolves that no pack member can harm another wolf's imprint, preventing internal conflicts. They can only imprinted on one person and one person only! It does not switch. It stays.
