According to Lauren Faust, each of the Supervillains is based on a lethal motivation similar to that of the seven deadly sins, with Harley Quinn's being chaos.
Harleen's civilian appearance is possibly based on Margot Robbie's portrayal in Suicide Squad.
This version of Harley Quinn has two hyenas named Ethel and Lucy instead of Bud and Lou.
Unlike other versions of Harley Quinn, this one doesn't have a romantic relationship with Poison Ivy. As for the Joker, Harley idolizes him. In the episode "The Warrior and the Fool," it's revealed that she's also in love with him.
In "Gotham Con", in the scene where Harleen tries to get rid of a bomb and throws it into a safe (only to discover it was the same safe the Joker had), she turns to the camera and says, "There are days when you just can't get rid of a bomb!", which is a reference to the 1966 Batman movie where Batman (played by Adam West) tries to get rid of a bomb on a pier, uttering the same phrase.
Like her Generation 1 counterpart, this version of Harley Quinn is not a psychiatrist, due to her young age.
She is the second member of the Supervillains to redeem herself; the first is Star Sapphire.
However, the difference lies in the fact that Star Sapphire has not been fully reformed, while Harley Quinn became a permanent part of the Superhero Girls in Mayhem in the Multiverse.
This is the first version of Harley that shows her as Barbara Gordon/Batgirl's best friend instead of Pamela Isley/Poison Ivy.
Interestingly, it seems that Babs is the only one who hasn't been a victim of Harley's pranks.
The relationship between Harleen and Barbara is similar to that of Peter Parker/Spider-Man and Harry Osborn/Green Goblin from Marvel Comics.

As Harleen, she's a fun-loving girl who enjoys playing pranks. She can be a bit of a handful, for example, when the other Super Hero Girls meet her. As Harley Quinn, she still enjoys pranks, but she can also show her mischievous side. In both cases, she can be vengeful, like when Robin made fun of Babs.

As Harley Quinn, she wears pale, almost white, face paint and thin black eyebrows. She wears a black mask and retains her red lipstick. She wears a two-tone jester's hood, red on the right side and black on the left, finished with white pom-poms. She wears a form-fitting, full-body jumpsuit with an asymmetrical, multicolored pattern. The colors alternate across the body: the right arm, left side of the chest, and right side of the lower torso down to the right leg are red, while the left arm, right side of the chest, and left side of the lower torso down to the left leg are black. On the outside of the right thigh, there is a distinctive three-diamond pattern in black. A prominent, oversized white collar with ruffles and white pom-poms surrounds her neck. She also wears matching white ruffles on her wrists. She wears mismatched gloves: red on the right hand and black on the left, matching the opposite color of her sleeves. Her shoes are pointed jester-style boots with curled toes, continuing the mismatched red and black color theme; her left boot is black, while her right boot is red.

Harleen is a tall, slender, fair-skinned teenager with rosy cheeks, sky-blue eyes, and long, light blonde hair styled in two high pigtails. The tips of her left pigtail are dyed blue, while those of her right pigtail are dyed red. She also has small blue and red streaks in her bangs. She wears red lipstick.

As a civilian, Harleen wears a varsity-style bomber jacket with a vertical vent down the center. The left side is red and the right is blue. Both sleeves have white stripes at the cuffs. She also wears matching shorts, also with a vertical vent, red on the left and blue on the right. The hems of the shorts appear frayed or ripped. She wears knee-high boots of different colors. The left boot is red with white details and red laces, and the right boot is blue with white details and blue laces.
