Family
Jeyne is the last direct survivor of her branch of House Arryn after the murder of her father and older brothers at the hands of the Stonecrows. Her cousin, Ser Arnold Arryn, contested her inheritance twice, which led Jeyne to disinherit him and his descendants entirely from the line of succession. Through her kinship with Aemma Arryn, Rhaenyra Targaryen's mother, Jeyne maintains a distant blood tie to House Targaryen, a connection that Rhaenyra will later attempt to exploit politically.

Early life
Jeyne became Lady of the Eyrie in 97 AC, when she was just three years old, after the Stone Ravens murdered her father and older brothers. Due to her young age, she did not attend the Great Council of 101 AC; instead, Lord Yorbert Royce served as Lord Protector and regent throughout her minority.
Her inheritance was contested twice by her cousin, Ser Arnold Arryn, who was even imprisoned in one of Heaven's dungeons for it; upon reaching her majority, Jeyne disinherited him and all his descendants in her will. Jeyne herself would later claim that her relatives had attempted to replace her "three times," although the chronicles only document two direct rebellions by Arnold; it is speculated that the third occasion she referred to was either when she was installed as ruler of the Vale at the age of three, or when she assumed full power in 110 AC.

After the death of Lady Rhea Royce in 115 AC, her widower, Prince Daemon Targaryen, approached Jeyne hoping she would recognize him as heir to Runestone. Jeyne not only flatly rejected his claim, but also openly warned him that his presence in the Vale of Arryn was unwelcome, thus setting the tone for her relationship with House Targaryen: cordial when convenient, but without gratuitous concessions.

Season 1
Following the suspicious death of Rhea Royce, Daemon proclaims at Rhaenyra and Laenor Velaryon's wedding that he will immediately fly to the Eyrie to ask Jeyne to name him heir to Runestone, since Rhea died childless. The attempt is unsuccessful, and Daemon ends up exiled in Pentos.
Years later, as tensions erupted leading up to the Dance of the Dragons, Rhaenyra was confident that Jeyne would side with her, thanks to their blood relation through Aemma Arryn. She sent her son Jacaerys to negotiate with her personally.
Season 2
Jacaerys successfully negotiates Jeyne's support. The Lady of the Eyrie commits the forces of the Vale to the Blacks' cause, but imposes a non-negotiable condition: that a dragon rider protect the Vale in return.
