Route 06 is a survival game for up to five players. Players control one of nine survivors, each with their own passive and/or active ability that can be used to their advantage and that of their teammates. If a player runs out of health, they will be defeated and their blood will drain. Players have the ability to revive defeated teammates under various circumstances until they run out of blood and die. At the beginning of the game, at the 0 mile mark, players have a 5-seater car and a baseball bat as their starting weapon. They must travel along a road known as Route 06 for at least 4000 miles[n 1] to a military base to receive help. The trunk of the car can hold a certain[n 2] number of items. Players will encounter various randomly generated structures, which typically contain valuable items that can be sold for money, as well as consumables and weapons. Players will progress through various biomes, beginning with the Plains—the introductory biome. Throughout the game, players will encounter zombies, the game's primary enemies. Zombies come in different types, and some are more resilient than others. They typically hide in structures or may emerge from the ground randomly. Players can defeat them with weapons, dealing a certain number of hits depending on the weapon used and the zombie type. They can also use a character's active ability.

At certain stages, players will reach Checkpoint C.4.RL, where they can take a break and sell their loot for cash, which can be spent on weapons and consumables. Once all players are prepared, they can move on to the next stage. After the first checkpoint, players will enter a swampy biome, where the Plague enters play. The Plague is an unkillable zombie that appears only at night. At nightfall, it will immediately begin stalking and attacking players.

Clayton is a cyborg with yellow skin, shaggy white hair, and a short white beard. Being partially robotic, many parts of his body are robotic, including the right side of his face, part of his left arm, his right leg, and his right hand, which has been replaced by his trusty railgun.

He wears an open dark purple jacket, a black tank top, and blue jeans with a noticeable rip in the leg. Clayton's past, like most of the other survivors, is largely unknown. It was revealed that his "Past Scars" skin is canon, meaning he served in the military in his youth and likely suffered serious injuries, leading to robotic enhancements and possible discharge. He also served alongside Marcus until he suffered an injury, for which Marcus blames himself. This is supported by his dialogue with Ethan, in which he says that before the infection, he "barely used his hand cannon."

Marcus is a grown man with yellow skin, a bald head, and a small, black beard on his lower jaw. For attire, he wears a millitary cap, stationed on his head. He has a light green shirt, with brown gloves and a bit of hair scattered on his arms. On the lower half of his body, he wears a brown belt with yellow buckles, camouflaged pants, and brown boots Prior to the events of the game, Marcus served as a soldier in the military. He has some history with Clayton, the two having served together. The latter character would soon sustain severe injures that Marcus blames himself for this Marcus always values ​​old friends and is the first to apologize. He's a confident, fearless man. He's ready to fight for his country and, most importantly, his family. He's moderately strict. He's better at hand-to-hand combat than marksmanship. He hates it when Clayton tries to protect everyone at the cost of his life. After the war, out of guilt, he tries to help Clayton everywhere, while calming him down in a commanding tone when he makes mistakes, eschewing the gray-haired man's fervor.
