She is an intelligence analyst for the Federal Bureau of Investigation (FBI). Eight years after the murder of her adoptive mother, Alyssa Ashcroft, Grace was targeted by Victor Gideon and Zeno, of the clandestine organization called The Connections, who believed Grace was the key to unlocking Elpis. Originally an orphan, Grace was adopted by one of Umbrella's founders, Oswell E. Spencer, whom she considered her blind hope and redemption for everything he had done after the 1998 Raccoon City incident. Wishing to leave a legacy and entrust Grace to safer hands, Spencer readily agreed to an interview request from investigative journalist and Raccoon City outbreak survivor Alyssa Ashcroft, in which he would give his side of the story, hoping it would serve as "a requiem for the dead." Toward the end of the interview, Alyssa was alarmed to see Spencer holding infant Grace, believing her to be one of his many test subjects in his quest for evolution and divinity. However, Spencer denied this, stating that Grace was a normal child he had taken in. Spencer asked Alyssa to hold little Grace and designated her as his blind hope. After Spencer's death in 2006, his butler, Patrick, wrote to Alyssa informing her of his master's will, in which he granted her guardianship of Grace and made her her adoptive mother. Eventually, Grace adopted her mother's surname, Ashcroft.

Due to the nature of her mother's freelance work, they moved frequently, going from place to place wherever Alyssa found employment. This inadvertently impacted Ashcroft's social skills, as the constant moves made it difficult for her to form friendships. In 2004, former Umbrella scientist and Spencer devotee Victor Gideon learned of Spencer's latest research on memory blood and its ability to transfer memories via the t-virus. Gideon believed Elpis was Spencer's final work, a powerful virus, but it was locked away by a password at ARK, a deep underground facility beneath the ruins of Raccoon City. Eventually, Gideon learned of Grace's existence and surmised that she was the successful infant clone born from the memory transfer experiment, possessing the necessary Spencer memories to unlock Elpis. In December of that year, Gideon obtained her incomplete medical records and attempted to model the many clones after her, but he was still unable to replicate Grace. While Gideon and the Connections were providing clones of ARK and trying to replicate it, he also tried to locate it.

Wrenwood Hotel Case (2018)

In 2018, Ashcroft and her mother arrived in the town of Wrenwood, where Ashcroft had found her next job. They stayed there for two days until her mother finished her work. However, while conducting research, Alyssa was stalked by a hooded man infected with a mutated strain of the T-virus, sent by Gideon as an assassin. That night, Ashcroft and her mother were staying at the Wrenwood Hotel when Ashcroft received a call from Gideon, who confirmed his identity before the call abruptly ended. Shortly afterward, the hotel experienced a power outage, and her mother, suspecting that the hooded man was coming for them, quickly convinced her to leave with her. Despite her confusion, Ashcroft followed her mother to the lobby. There, the manager of the Wrenwood Hotel assured her that the power outage was normal due to the hotel's 40-year-old electrical system, before being murdered by the hooded man, who slit his throat. Her mother, Alyssa, quickly shot the man and locked him behind the back door of the reception area. Ashcroft was shocked by her mother's actions and saw her hide her purse behind the painting. She tried to ask what was in it, but was told she would explain later. Upon reaching the emergency exit, Ashcroft suffered a panic attack after calling the local police to report the murders, despite her mother's protests. Her mother reassured her, reminding her that she was "her hope," though Ashcroft didn't understand her words. They tried to leave, but Ashcroft saw the hooded man slash her mother's throat with a machete.
