These are three extremely difficult tasks described on one of the "Tablets of God" (ancient stone tablets). If a person completes all three trials and recites a final incantation, they will close the Gates of Hell forever, trapping all demons inside and preventing them from returning to Earth. Sam is the one who decides to do them, which initiates a mystical purification process in his body, but also a serious physical deterioration. The 3 Trials (What Sam Must Do):

-Kill a Hellhound and bathe in its blood: Sam manages to kill an invisible one that was stalking a family and, after bathing in its blood, recites the first incantation. From this point on, his hands begin to glow at times and he starts to feel dizzy.

-Rescuing an innocent soul from Hell and taking it to Heaven: With the help of a reaper, Sam physically travels to Purgatory and then to Hell. -Curing a demon: The final test involves injecting purified human blood (blood from someone who has confessed or is "good") into a demon every hour for eight hours, until the demon regains its humanity. Sam attempts to do this with Crowley, the demon king of Hell.

He doesn't complete them, because before he finishes the third one, Dean forces him to stop, explaining that he will die if he completes them. Even so, Sam felt that this was his purification, a small redemption after all the sins he has committed.
