Chizome is an intimidating, muscular man with a flat, somewhat triangular face, no nose (which he removed himself), and an unusually long tongue covered with numerous small papillae. He has rather long black hair, unkempt except for the headband and mask, and small eyes with tiny red irises.

As the Spot, he wears a dark battlesuit covered in metal armor over his entire body for weapons and protection. He wears a black undershirt, and his arms are exposed, so he wraps them in bandages from just below his shoulders to his wrists. After graduating from high school, the teenage Chizome preached an ideology he called "the revival of Heroica" through street oratory and various speeches, but his words went unheard. Over the next decade, Chizome realized that only action could create his ideal world and change Heroica for the better. Chizome researched and disciplined himself, training himself in the art of assassination through self-study, to achieve his duty as the "Revival of Heroica." During this decade, Chizome's parents died. The cause of their deaths is unknown, but it is believed Chizome was not involved.

Stain can paralyze his target by ingesting samples of their blood. The amount of time the victim is paralyzed varies depending on their blood type, with B having the longest duration, followed in descending order by AB, A, and O. Regardless, the maximum amount of time he can be paralyzed is eight minutes. Stain uses blades to draw blood from his opponents to utilize his Quirks. Previously, the Hero Killer deeply desired to become a hero, but during his training, he disliked the principles of modern heroes.
