Leon already shows himself to be a man who protects at any cost, especially children. After the events of Raccoon City, Sherry falls under government control. Sherry and Jake reach the meeting place with Simmons, who is talking with Leon and Helena. Sherry realizes that Simmons is indeed involved in the president's death, after which a firefight ensues between the agents and Simmons' men. During the battle, Sherry and Jake are unexpectedly attacked by a group of J'avo, who take them captive. Waking up, Sherry apologizes to Jake for trusting Simmons. Suddenly, they are freed. The complex begins to crumble, but Jake and Sherry manage to find a platform on which they begin their journey toward the exit. Along the way, a seriously wounded and burned Ustanak hinders the heroes once again, but Jake and Sherry shoot the mutant in the heart, killing him completely. Jake thanks Sherry for saving his life. The heroes then exit the complex on the platform, which soon explodes. Afterward, Sherry resumed her work with the DSO.

The partners search for a way out, but encounter Ustanak again. They escape from him, then defeat him by piercing his stomach with a drilling rig. While escaping the cave, Jake and Sherry are ambushed by rebels and then unexpectedly attacked by Ustanak, who backhands Sherry, knocking her unconscious. A second later, a woman in blue appears—Ada Wong (who was actually Carla Radames). She tells Jake that he is Albert Wesker's son and that he has special blood, passed down from his father. After telling her story, Ustanak knocks Jake unconscious. Six months later, Jake and Sherry are being held in a secret Neo-Umbrella research facility in China for experiments. At one point, Sherry and Jake manage to neutralize the guards and escape the cell. After destroying numerous J'avo, they escape the facility. Due to the J'avo's numerous forces and a pursuing helicopter, Jake and Sherry's escape routes were cut off. With the help of Chris Redfield and other BSAA members, they repelled the terrorists' attack and soon destroyed the enemy helicopter. Along the way, the partners witnessed a plane crash, and Sherry noticed that the plane was American. They soon spotted two American agents—Leon Kennedy and Helena Harper. After a brief conversation, Leon revealed that Simmons was the mastermind behind the crash. Shocked by what he heard, Sherry couldn't believe it, as Simmons was her direct superior. Tensions escalated when Leon pressured Sherry, to which Jake immediately reacted and pushed him away. Their conversation was soon interrupted by Ustanak. They all managed to repel the attack and parted ways. Later, the heroes encounter a BOO called Ubisto, who has emerged from a cocoon and nearly kills Sherry, but the real Ada Wong suddenly appears, saves Sherry and helps destroy the mutant, after which she escapes.

In December 2012, Derek Simmons, then National Security Advisor, ordered Sherry to Edonia in search of the mercenary Jake Muller. It was discovered that his blood contained antibodies that could help fight a new virus, the C-virus. In the opening cutscene, rebels are seen injecting themselves with the virus and showing the first signs of mutation. One of the rebels, mercenary Jake Muller, also injects himself with the virus, but shows no signs of mutation. Sherry immediately identifies the mercenary, and they both escape. While killing J'avo along the way, Sherry tells Jake that the world needs his blood to create a vaccine for the C-virus. Jake reluctantly agrees, but only on the condition that he be paid $50 million in cash. Soon, the partners are pursued by Ustanak, a powerful bio-organic weapon. After a brief fight in an old building, Sherry and Jake fall through the floor. Jake and Sherry soon met up with BSAA operatives Chris Redfield, Piers Nivans, and others. After destroying the anti-aircraft guns and the B.O.W., Jake and Sherry were evacuated by helicopter on Captain Redfield's orders. After a brief conversation between Jake and Sherry, the helicopter is attacked by Neo-Umbrella and Ustanak, who destroy the BSAA helicopters. At the last moment, Jake manages to grab the parachute device, but the parachute is damaged, and both fall into the snowy mountains. The partners manage to survive, but Jake notices that Sherry is seriously injured. Sherry persuades Jake to remove the shard. Jake removes the shard and is amazed by what he sees: Sherry's massive wound begins to heal instantly and, after a few seconds, is completely healed. Hiding in a cabin from the blizzard and the J'avo patrolling the mountains, Sherry tells Jake about what happened in Raccoon City 15 years ago and about her father, who invented the G-virus. Sherry was one of the few survivors in the city and the only one to adapt the virus, granting her the ability to regenerate.

Sherry and Claire tried to escape together. They managed to reach the sewers, where the mutated William was still trapped. He sensed his daughter's presence and began pursuing her. When Claire and Sherry separated, her father found his daughter and implanted the G-virus embryo into her. However, due to her blood relationship, the girl did not die instantly, as had happened to other people. After finding Sherry, Claire discovered a way into Umbrella's underground lab and went there with her. There, she put the girl to bed in the security room and began searching for information on how to save her. Claire later met Annette, who told her about a vaccine against the G-virus called "Devil." Having successfully created the vaccine, Claire injected it into Sherry, saving her life, although the changes to her genome were already irreversible. Annette herself was subsequently killed by her husband. Leon, Claire, and Sherry soon boarded the laboratory's emergency train, but William caught up with them, clinging to the rear cars. Leon and Claire managed to shake him off, and the three of them escaped the complex before it self-destructed. After escaping Raccoon City, Kennedy insisted that Claire continue her search for her brother. After she left, Leon and Sherry were found by the authorities. The girl was placed under guard and kept under surveillance. Leon subsequently managed to guarantee Sherry's safety, but at the cost of being forced to join a top-secret military agency. In addition to the military's research interest in Sherry, Albert Wesker also posed a serious threat. Sherry's legal guardian was Derek K. Simmons, who planned to use her for private research after the closure of the American bioweapons project. During her years in custody, Simmons' team, using regular blood tests to analyze Sherry's blood, managed to isolate and create the C-virus. After Wesker's death in 2009, Sherry became a government agent.

Due to William's dissatisfaction with his position at the company, he decided to hand over his research to the Pentagon in exchange for protection for himself and his family and the right to continue research on the G-virus, which he agreed to. Umbrella learned of the betrayal and, to forestall the military, sent a security team to his lab on September 22 to seize samples of the G-virus. During the raid, the team shot William and stole his samples. The seriously wounded scientist, in retaliation, infected himself with the remaining sample, transformed into a monster, and killed almost all of his attackers, inadvertently spreading the T-virus to rats, who then contaminated Raccoon City's drinking water.

On September 24, tens of thousands of city residents turned into zombies. A worried Annette contacted Sherry, who was at school at the time, and told her to go home immediately. At home, her mother called again and told her to get a necklace from the box in her bedroom and then go to the police station, which was supposed to be safe. At the police station, Sherry called her mother from the phone in the hallway, reporting that the building was empty and that she had heard the monster growling. Annette then ordered her to go to the second floor and find a place to hide. She did so. On September 29th, Sherry met Claire Redfield at the police station, who had come to town searching for her missing brother, Chris. Also in the building was newly arrived rookie police officer Leon Kennedy. He only saw Sherry once, but that was enough to suppress his urge to commit suicide.

Sherry was born in 1985 or 1986 to William and Annette Birkin, two virologists involved in T-virus research at a secret lab in Arklay County. Due to her parents' busy work schedules, Sherry was often neglected, even when they were transferred to an underground lab in Raccoon City in the early 1990s. Her parents were senior researchers on the G-virus project, which they believed would improve the human race.
