Sherry and Jake reach the meeting place with Simmons, who is talking with Leon and Helena. Sherry realizes that Simmons is indeed involved in the president's death, after which a firefight ensues between the agents and Simmons' men. During the battle, Sherry and Jake are unexpectedly attacked by a group of J'avo, who take them captive. Waking up, Sherry apologizes to Jake for trusting Simmons. Suddenly, they are freed. Killing numerous Rasklapanya mutants, they manage to activate the elevator, but are nearly killed by the overload. Along the way, they encounter Chris and Pierce. Chris personally tells Jake that he was the one who killed Albert Wesker. Jake becomes enraged and nearly kills Chris, but restrains himself in time. At this moment, a gigantic B.O.W. Chaos emerges from its cocoon, designed to infect and destroy the entire world. Chris and Pierce remain behind to fight Chaos, while Sherry and Jake head for the complex's exit. On their way out, Sherry and Jake encounter Ustanak again. After a long, grueling battle, Jake delivers a powerful punch that sends Ustanak flying into the lava. The complex begins to crumble, but Jake and Sherry manage to find a platform on which they begin their journey toward the exit. Along the way, a seriously wounded and burned Ustanak hinders the heroes once again, but Jake and Sherry shoot the mutant in the heart, killing him completely. Jake thanks Sherry for saving his life. The heroes then exit the complex on the platform, which soon explodes. Afterward, Sherry resumed her work with the DSO.

The partners search for a way out, but encounter Ustanak again. They escape from him, then defeat him by piercing his stomach with a drilling rig. While escaping the cave, Jake and Sherry are ambushed by rebels and then unexpectedly attacked by Ustanak, who backhands Sherry, knocking her unconscious. A second later, a woman in blue appears—Ada Wong (who was actually Carla Radames). She tells Jake that he is Albert Wesker's son and that he has special blood, passed down from his father. After telling her story, Ustanak knocks Jake unconscious. Six months later, Jake and Sherry are being held in a secret Neo-Umbrella research facility in China for experiments. At one point, Sherry and Jake manage to neutralize the guards and escape the cell. After destroying numerous J'avo, they escape the facility. Due to the J'avo's numerous forces and a pursuing helicopter, Jake and Sherry's escape routes were cut off. With the help of Chris Redfield and other BSAA members, they repelled the terrorists' attack and soon destroyed the enemy helicopter. Along the way, the partners witnessed a plane crash, and Sherry noticed that the plane was American. They soon spotted two American agents—Leon Kennedy and Helena Harper. After a brief conversation, Leon revealed that Simmons was the mastermind behind the crash. Shocked by what he heard, Sherry couldn't believe it, as Simmons was her direct superior. Tensions escalated when Leon pressured Sherry, to which Jake immediately reacted and pushed him away. Their conversation was soon interrupted by Ustanak. They all managed to repel the attack and parted ways. Later, the heroes encounter a BOO called Ubisto, who has emerged from a cocoon and nearly kills Sherry, but the real Ada Wong suddenly appears, saves Sherry and helps destroy the mutant, after which she escapes.

In December 2012, Derek Simmons, then National Security Advisor, ordered Sherry to Edonia in search of the mercenary Jake Muller. It was discovered that his blood contained antibodies that could help fight a new virus, the C-virus. In the opening cutscene, rebels are seen injecting themselves with the virus and showing the first signs of mutation. One of the rebels, mercenary Jake Muller, also injects himself with the virus, but shows no signs of mutation. Sherry immediately identifies the mercenary, and they both escape.
