Ashley Graham is the daughter of the President of the United States. Prior to the events of the game, she is kidnapped by a mysterious group in rural Europe. The government discovers that the culprit is a sect called “Los Iluminados,” which worships a parasite known as Las Plagas. Agent Leon S. Kennedy, a survivor of the Raccoon City incident, is sent alone to rescue her.

During the rescue, Leon discovers that Ashley has been infected by the parasites, making her not just a victim but a potential tool of the cult. The mission then becomes not only to save her but also to rid her of the infection and prevent the group from using the plague to take over the world.

Throughout their journey, Ashley and Leon develop a bond of trust and friendship. She goes from a scared and powerless young woman to a courageous, empathetic, and determined character, facing dangerous situations alongside him.

The remake emphasizes her emotional growth — Ashley goes from being just “the girl who needs saving” to someone who wants to fight for her own survival.

Ashley is kidnapped on her way home from college and taken to a secluded village in Spain. There, cult leader Osmund Saddler orders her to be injected with the Las Plagas parasite, with the plan to control the US president through his daughter.

Los Iluminados' goal is to send Ashley back to the U.S. infected—the parasite would mature and allow Saddler to control both her and the president, spreading his influence globally. Leon arrives at the village after receiving information about Ashley's whereabouts. After battling the infected villagers (ganados), he finally finds her in a church.

When the two reunite, Ashley is terrified but still lucid. Leon promises to get her out and protect her. From then on, the two work together, escaping hordes of enemies, navigating traps, and trying to survive. As the parasite grows inside her, Ashley begins to experience seizures, fever, and intense pain, at times nearly losing control. This shows that time is running out—if the parasite matures, she will be completely overwhelmed. These moments also reveal her mental strength: even in despair, Ashley tries to resist and help Leon, showing that she doesn't want to be a burden, but someone who is also part of the fight. During their escape, Ashley and Leon arrive at the castle of the nobleman Ramon Salazar, an ally of Saddler. There, Ashley is separated from Leon several times — one of these occasions leads to one of the most memorable parts of the remake, where the player controls Ashley directly. In this section, she's alone, weaponless, armed only with a flashlight. She must solve puzzles and escape from animated suits of armor controlled by the parasite. Further on, Leon and Ashley meet scientist Luis Serra, a former Umbrella researcher who is familiar with the Las Plagas parasite. At the end of the game, both undergo a process of removing the Plagas — an extremely painful procedure, but one that frees them from the cult's control.
