After being captured by HYDRA, he was used as a test subject by Arnim Zola, who administered a prototype of the Super Soldier Serum. Although he managed to escape with Steve, months later he fell from a train during a mission in the Alps. Everyone presumed him dead.

HYDRA recovered his body. He had survived thanks to the serum, but he had lost his left arm and was severely injured. Instead of letting him die, the organization rebuilt him. They replaced his arm with a high-performance titanium prosthesis and began a decades-long process of physical and psychological torture. He was subjected to constant brainwashing, memory erasure, hypnosis, and mind conditioning, gradually eliminating any trace of James Buchanan Barnes until he was nothing more than a weapon.

Thus the Winter Soldier was born.
