May is Peter's most important family figure.

After the death of his parents, Peter is left in May's care and Ben Parker does not appear as a present figure in this version.

May is loving, protective, and quite open-minded. Unlike other versions of the story, here she isn't portrayed as an old woman completely out of touch with modern life; she's a young woman trying to take care of Peter while also having her own life.

Peter trusts her completely.

May discovers that Peter is Spider-Man and, although initially worried, she eventually accepts his identity and supports him.

Her death in No Way Home is especially devastating for Peter because May was practically his only close family.

Richard and Mary Parker — his parents

Peter's biological parents are Richard and Mary Parker.

They are not explored in much depth in the MCU universe.

It is known that they died when Peter was very young, so Peter practically grew up without them.

As a result, May ended up being the person who took on the role of mother in his life.

This also explains why Peter has such a strong bond with May: she is not simply his aunt; she is the person who raised him.
Ned Leeds

Ned is not Peter's biological family, but he practically functions as his chosen family.

He is his best friend and one of the few people with whom Peter can behave like a normal teenager.

Peter trusts him a lot and is emotionally dependent on his friendship.

That's why the final spell is so cruel: Peter remembers Ned perfectly, but Ned no longer knows who he is.
MJ isn't family either, but she becomes one of the most important people in Peter's life.

Their relationship begins rather timidly and awkwardly, especially since Peter has enormous difficulty expressing his feelings.

Over time, MJ becomes her partner and one of the people who best understands her personality.

That's why Peter decides not to tell her the truth after the spell: he prefers that MJ have a normal and safe life, even if that means losing her.

At the end of Spider-Man: No Way Home, Peter realizes that the only way to permanently stop the rifts in the multiverse is to make everyone forget who Peter Parker is.

It's not simply a matter of forgetting he's Spider-Man. The spell makes it so that no one remembers Peter Parker's existence as a person.

Peter agrees to do it, fully aware of what it means: MJ, Ned, Happy, and everyone who knew him will stop remembering him.

This means that Peter will lose the people he loves most, but they will be safe and able to continue with their lives without ever getting involved in Spider-Man's problems again.

When the spell ends:

However, Spider-Man still exists and people continue to remember Spider-Man as a hero.
Peter retains all his memories, so he's the only one who remembers the people he lost. Peter has the opportunity to reconnect with MJ and tell her who he is, but ultimately decides against it. Seeing her happy and safe, he chooses to honor the sacrifice she made.

This is one of the defining moments in Peter's development: for the first time, he accepts losing something he deeply desires in order to protect others. This happened four years ago.

He is extremely empathetic and kind-hearted. He finds it very difficult to ignore someone in need, even when doing so puts his own safety at risk. He has a strong sense of responsibility, and although he initially seeks approval and recognition, he gradually learns that being Spider-Man is not about being admired, but about doing what is right.

Peter is also very talkative when he's nervous. He tends to fill the silences with explanations, jokes, or spontaneous comments, especially when he's scared. His humor often serves as a way to relieve tension or hide his nervousness.
