Gwayne arrived at court in 101 AC with his siblings, mother, and father when his father was appointed Hand of the King to King Jaehaerys I Targaryen. In 111 AC, Ser Gwayne participated in a tournament in King's Landing to commemorate the fifth anniversary of King Viserys I Targaryen's marriage to Gwayne's sister, Queen Alicent Hightower. Gwayne was one of his sister's champions during the tournament but was unhorsed by Ser Criston Cole. When the Dance of the Dragons began in 129 AC, Gwayne was appointed second-in-command of the City Watch of King's Landing by his father, who instructed him to keep an eye on Ser Luthor Largent, the Commander of the City Watch, for any signs of disloyalty to the newly crowned King Aegon II Targaryen. Gwayne and Ser Arryk Cargyll were in Grand Maester Orwyle's entourage when he traveled from King's Landing to Dragonstone in an unsuccessful attempt to reconcile Aegon II and his half-sister, Rhaenyra Targaryen. When King's Landing was attacked by the Blacks in 130 AC, Gwayne rushed to the stables to sound the alarm, but was seized, disarmed, and dragged to their commander, Luthor Largent, who was still loyal to Prince Daemon Targaryen. When Gwayne denounced him as a turncoat, Ser Luthor plunged his sword into Hightower's belly and ordered the city gates opened to the Blacks.
