She wears large glasses, a green turtleneck sweater, dark pink trousers, and brown shoes without socks, which dangle to illustrate her impassive expression. And as stated and shown in "Potty Mouth," she is bald, toothless, and has an extra toe on her right foot due to her experiments, which forces her to wear a short, shaggy brown wig and false teeth to appear normal.

From the fifth season onwards, she is now occasionally depicted with permanent eyebrows when expressing emotions with her eyes.

Her sleepwear consists of a lime green nightgown with three white buttons and matching trousers.

Her swimsuit consists of blue sunglasses and a green one-piece swimsuit with a horizontal pink line down the center.

Although she's the second youngest of the Louds, Lisa is a child prodigy, making her the smartest of the group. Because of this, she's often self-centered, rude, condescending, and sarcastic with others. She speaks with a lisp and usually has a frown on her face, though she does smile occasionally.

As she herself states in "Heavy Meddle," she doesn't usually care about "mere human emotions," but she does get excited when something really good happens, such as when she and the other sisters discover Lincoln's romantic situation in "Heavy Meddle." She was also shown to have a crush on Hugh, just like all her sisters, and displayed a more immature side, even going so far as to make Hugh show off his abs by lying that she was teaching Lola human anatomy as an example. She also became somewhat irrational, stating that she would marry Hugh and have children with him, despite the obvious age difference, and following him everywhere like her other sisters. She also doesn't believe in superstitions, divination, blessings, or curses, as she is a realist and only believes in science, as stated in "Raw Deal" and "Study Muffin." However, she's not entirely close-minded, as she believes in aliens, as she stated in "Friend or Fake?", and accepts Luna dating a girl, as seen in the comic "Sam's Choice". Even so, she enjoys spending time with her siblings and having fun with them, though she doesn't show it.

In many episodes, she can be seen working on strange experiments. In "Left in the Dark," she creates an Oreo-shaped cookie called "Gloweos" using jellyfish DNA, and if someone were to eat it, their entire body would glow. One of her favorite pastimes is solving very difficult math problems. She also enjoys studying her siblings' feces and bathroom habits, something they all find disgusting and annoying.
