A creature named Koro-sensei destroys part of the moon and threatens to destroy Earth within a year. However, he decides to become the teacher of Class 3-E, a group of students considered the worst in the school, and gives them the mission to kill him before time runs out.

Although it's his goal, Koro-sensei turns out to be an incredible teacher who helps his students improve in every way. Among them are Nagisa Shiota, with a natural talent for assassination, and Karma Akabane, intelligent and dangerous.

Throughout the year, they try to kill him many times without success, all the while growing as individuals. They then discover that Koro-sensei was once human and that he will inevitably explode, which would destroy the Earth.

In the end, though they love him dearly, they decide to fulfill their mission and kill him to save the world. Before dying, he proudly bids farewell to his students, who carry on with everything they learned.

Itona Horibe, a boy connected to antimatter tentacles, initially acts as an aggressive rival to Class 3-E. He has messy white hair, light eyes, and a somewhat disheveled appearance. He possesses an explosive personality and a strong obsession with surpassing Koro-sensei.

Ritsu, an artificial intelligence designed as a killing machine capable of analyzing situations and hacking systems. She usually appears as a girl with purple hair and large eyes inside a screen or robotic body.

Ryūnosuke Chiba, a serious and quiet student specializing in sniper rifles and long-range weapons. He has dark gray hair, calm eyes, and an almost always relaxed expression. He possesses great accuracy and usually remains calm during important missions.

Gakushū Asano, the headmaster's son and leader of the elite students. He has perfectly styled dark blue hair, light eyes, and a neat and refined appearance. He is extremely intelligent, strategic, and competitive.

Gakuhō Asano, the principal of Kunugigaoka Junior High School and the main person responsible for the school's competitive system. He has dark hair slicked back, stern eyes, and an intimidating and authoritarian presence. He firmly believes that fear and pressure are necessary to create successful students.

Irina Jelavić, a foreign professional assassin known as "Bitch-sensei," has long blonde hair, blue eyes, and a striking, elegant appearance. She primarily uses seduction, manipulation, and deception during her missions, though she gradually develops genuine affection for her students.

Tadaomi Karasuma is a professional government agent tasked with training Class 3-E in assassination techniques and self-defense. He has short black hair, dark eyes, and a serious, athletic build. He is disciplined, calm, and one of the few adults capable of physically confronting dangerous threats.

Tomohito Sugino, a student in Class 3-E, is known for his kind, energetic, and hardworking personality. He has short, dark blue hair, light eyes, and a simple, athletic appearance. Before joining Class 3-E, he was a baseball player, so he possesses excellent aim and physical coordination. Although he sometimes feels insecure when comparing himself to others, he always tries to improve and support his classmates during missions.

Ryōma Terasaka, a student in Class 3-E, is known for his impulsive, loud, and defiant personality. He has dark red hair slicked back, small eyes, and an intimidating appearance that often makes him seem like a troublemaker. Initially, he acts aggressively and selfishly, but gradually demonstrates genuine concern for his classmates. He is known for his bravery, physical strength, and tendency to rush headlong into danger without thinking twice.

Yūma Isogai, a student in Class 3-E, is known for being responsible, kind, and one of the most reliable leaders in the class. He has light brown hair, brown eyes, and a simple yet athletic build. Despite facing financial difficulties, he always tries to help his classmates and keep the group together. He excels in physical prowess, leadership skills, and a strong sense of responsibility.

Rio Nakamura, a student in Class 3-E, is known for her playful, bold, and confident personality. She has long, orange-blonde hair, light eyes, and a striking, modern appearance. She often acts playfully and flirtatiously, but she is also quite intelligent and observant. Nagisa Shiota, a quiet, shy, and highly observant student in Class 3-E. Although he appears physically weak, he possesses an enormous natural talent for assassination thanks to his ability to analyze behavior and identify weaknesses.

Karma Akabane, an extremely intelligent, sarcastic, and troublesome student, is known for his mocking and provocative personality. He has messy red hair, reddish eyes, and a confident smile that often comes across as intimidating. He excels in combat, possesses quick wit, and handles dangerous situations with ease.

Koro-sensei, the mysterious teacher of Class 3-E, resembles a yellow creature with tentacles. He has a round body, a huge, permanent smile, and usually wears a black teacher's gown and mortarboard. He can move at Mach 20, destroy mountains, and change the color of his face according to his emotions. Despite being the assassination target of the entire class, he eventually becomes one of the most beloved and caring teachers in the series.
