Like his siblings, Leonardo began as a baby turtle used for scientific research in Eric Sacks's laboratory. He was regularly injected with mutagen that gave him enhanced strength and healing abilities.

After April O'Neil freed the Teenage Mutant Ninja Turtles and Splinter, they began to grow rapidly in size and intelligence, comparable to or even surpassing that of humans. For the next fifteen years, Leonardo spent most of his life in the sewers with his brothers and adoptive father, where he learned the art of ninjutsu. His weapon of choice was the katana, with which he demonstrated great versatility (he was even able to cut a pizza with it in mid-air).

From the beginning of his life, he maintained a remarkably closer relationship with Splinter than with his brothers, often calling him "Dad" rather than simply "Sensei." Thanks to his greater maturity and sense of responsibility, Leonardo was appointed leader of the team, though this often led to clashes with Raphael.

Along with his brothers, Leonardo fought and defeated a squad of Foot Clan warriors who were about to execute hostages in the subway. He did so despite his reservations about disobeying Splinter. When they were found and photographed by an adult April O'Neil, Leonardo intervened to prevent Raphael from unduly bullying her, but still vaguely threatened to find her if she told anyone about them.

Leonardo had limited success fighting Shredder individually, but he led a team attack that allowed them to overwhelm the armored villain. When Shredder destabilized the toxin-filled tower, Leonardo ordered them to use their bodies to prevent it from collapsing. They were unsuccessful and barely survived the fall.

After recovering some of the mutagen, he and the other Turtles returned home and were able to cure Splinter. Later, he reunited with April, and they were calmer and more relaxed now that they knew she wouldn't reveal their secret.

Upon learning what had happened, Splinter ordered the Teenage Mutant Ninja Turtles to take April back to their lair, where Leonardo was shocked to learn of April's involvement in his origins. However, they were interrupted by an attack from the Foot Clan, which April had inadvertently led there, and Leonardo fought to defend his home and family. He was repeatedly electrocuted by the Foot Clan and forced to surrender when Shredder threatened Splinter's life. From there, he was captured along with Donatello and Michelangelo and taken to Sacks' mansion.

Along with Donatello and Michelangelo, Leonardo nearly died when his blood was drawn to collect the mutagen, but he managed to escape his cage when April administered a massive dose of adrenaline to the three Turtles. Overflowing with energy, they reunited with Raphael and set out to stop Shredder from detonating a chemical weapon in the heart of New York City. However, they were hampered by another attack from the Foot Clan, in which Leonardo was electrocuted again, rendered unconscious, and ultimately plummeted off a cliff.

Two years later, Leonardo clashed with Michelangelo and Raphael over the secrecy surrounding their lives. While Raphael resented his diminished status and Michelangelo longed to be part of the human world, Leonardo tried to remind them that it would never accept them and that their place was in the shadows as ninjas.

With Shredder's escape, along with his new alliance with Baxter Stogman and the Krang, a purple substance was discovered. April recovered a sample, and it was later observed that this substance functioned as a mutagen, creating new mutants under Shredder's control. Donnie discovered that with the right formula and enough of it, he could turn them into humans. Leo denied it and forbade Donatello from telling the others. But Mikey listened silently.

After Michelangelo told Raphael, another fight broke out between the brothers. Raphael was furious that Leonardo was withholding such vital information, while Leonardo insisted they were Turtles and would remain so. He left Michelangelo and Raphael behind while he and Donatello investigated a robbery at the museum and discovered that Shredder was planning to use a device to open a portal to another dimension. On his way home, he received a distress call from Michelangelo, and they tried to intervene at the police station, but ended up being exposed to the police.

Angry that Raphael had taken such a risk and gotten April into trouble, Leonardo vented to Splinter, telling him he couldn't get the Turtles' disparate personalities to work together. Although Splinter told him their different personalities were strengths of the team, his frustration grew after their failed mission in Brazil, culminating in him declaring that they were brothers, but not a team.

Upon returning to New York, the Turtles faced a difficult decision, where the purple slime and the transformation into humans seemed to be the only way to stop the Technodrome. Leonardo deferred to his brothers, saying he would agree to whatever decision they made.

Raphael destroyed the purple slime, reaffirming that they were and would remain Turtles.

In contrast, the Teenage Mutant Ninja Turtles offered an alliance to Chief Rebecca Vincent to prevent Krang from building the Technodrome over New York City and taking over the world. Leonardo fought Krang alongside his brothers, joining forces with Raphael to rescue Michelangelo from the alien's formidable robotic body. Their efforts resulted in the Technodrome and Krang being sent back through the dimensional portal.

Along with his brothers, Leonardo received a key to the city in front of the New York Police Department, but he decided to keep it a secret. He told Chief Vincent that April could contact them if the police needed them again.

Immense Strength: Leonardo is incredibly powerful thanks to the effects of the mutagen and his intense ninjutsu training. He is capable of defeating multiple Foot Clan ninjas in hand-to-hand combat.

Immense Speed: Leonardo is also extremely agile thanks to the Mutagen and his Ninjutsu training. His agility is incredibly high, capable of outrunning several Foot Clan ninjas and even keeping up with Shredder.

Master swordsman: Leonardo is a master ninja who is very proficient in the handling of his katana swords.

Mutated armored shell: Like his brother Leonardo, his naturally hard tortoise shell was mutated by the mutagen to grow in both size and density and has immense hardness that gives him bulletproof durability capable of protecting him against the barrage of rapid-fire machine gun bullets.

Katanas: Leo uses two katanas as his signature weapons, with a blue coating on the handle. Two years later, he uses the same katanas, but with a darker blue coating and additional coating on the end of each hilt.

Stealth: Thanks to his ninjutsu training, Leonardo is able to hide in the shadows and move through them without being seen or detected.

Foldable in its shell: Like the rest of the turtles, it is able to fold into its shell.

Expert marksman: Leo has proven to be an expert marksman, as he was able to throw one of his katanas at Shredder, knocking him off the giant beam with ease.

Indomitable Will: Like the rest of his brothers, Leo possesses an indomitable will, being able to endure 11 full hours in the Ha-shi, while balancing both legs on cement blocks, while using chopsticks to balance an egg in both hands and balancing a third egg on his head.

Leonardo has a faded scar that runs from his right forehead to his right cheek. He wears wooden chest armor, similar to the plates of a Yoroi samurai armor.

He also wears shoulder pads and a samurai skirt, as well as custom-made Japanese footwear. There are Japanese characters painted in blue on the back of Leo's shell.

Being the eldest, most cautious, and wisest of his brothers, he is a mutant turtle who uses a blue bandana and katanas as his signature weapons.
