Typically, Stein is a hedonistic scientist whose greatest pleasure is dissection and observation, which persisted from childhood to adulthood.[10] Due to this, Stein claims sees everything in the world (himself included), as an experiment. He developed sadistic tendies (due to his sensitivity to madness), has a high intelligence, and a driven personality.[10] His sadistic tendies can be seen during his battle against Maka and Black☆Star during the remedial lessons, in which he grabbed Maka by one of her pigtails, lifted her shirt up and drew marks on her body to signify where he would vivisect her and ponders if he should turn her skin into sand paper. Seeing as he was acting then and deliberately playing up his "mad doctor" persona for the sake of the lesson though, it's likely he didn't really mean it. He has shown a tendency to enjoy messing with his students, however. When Black☆Star tried to attack him in the same battle, he countered by knocking him out with his soul wavelength in a painful manner that caused Black☆Star to scream and convulse as though he were being electrocuted. His smiles typically are also full of sadism and serve to highlight the latent madness inside of him.[3]

During his childhood and adolescence, he had a tendency to want to dissect everything. He was stopped by his first weapon partner, Spirit Albarn.[10][11] He was shown to have little empathy even towards his friend/partner, having no qualms about secretly experimenting on Spirit's body. This eventually led to the decade-long dissolvement of their meister/weapon partnership after only five years.

Stein appears to be a lean but muscular male with silver-gray hair, large round glasses, and a handsome face according to some of his female students. He is likely in his late twenties or early thirties, seeing as he appears to be close in age to Spirit but is still a little bit younger than him. Stein's most notable trait is the large screw going through his head. He often turns this screw, as a means to focus his thoughts or as he prepares to attack an opponent.

Another notable detail about Stein's body, as well as his clothes, are the zig-zag stitches he has on both of them. He has stitches all over his body indicating experimentation on himself, and stitches all over his clothing. Even his house/lab is shown to be covered in stitches. His most notable piece of clothing is his white lab coat. He has a turtleneck shirt underneath that has a two different shades of gray pattern. The shirt also has zig-zag stitches.

Dr. Franken Stein , is an eccentric but talented doctor and meister who is known to be the strongest meister to have graduated from Death Weapon Meister Academy and formerly Spirit Albarn's first weapon-partner.[9] He later becomes a teacher at the academy upon Death's requestand was later assigned to act as a meister to Marie Mjolnir. Stein is loosely based upon the two central characters from the 1818 novel by Mary Shelley, Frankenstein, or The Modern Prometheus and its 1931 movie adaption, being an amalgamation of the mad scientist Victor Frankenstein and his nameless man-made monster. Like Victor Frankenstein, Stein has a mindset in which he wishes to live in a world "without God," setting his own rules rather than following those of some higher power. In addition, Stein also has motifs centered around electricity, similar to that used by Victor Frankenstein in the 1931 movie to give life to the creature. Both his attacks evoke electricity and his current partner has attacks centered around electricity. The screw in Stein's head resembles the bolts found in the head or neck in many popular portrayals of Frankenstein's monster that all follow the 1931 movie's lead. Stein has more moves than just electricity; he can also use his soul as a shield, as shown when Stein battled Crona. When he battled Medusa, he also uses an attack straight to Medusa's stomach, causing her to nearly die.

During his early childhood in kindergarten, Stein saw kids his age play together like they were to told to, an act which sickened him because he saw it as blind obedience. He also made an issue out of the children focusing on taking notes and following instructions given to them by the teacher; saying it made him feel like losing control.[48] Having a disposition towards violence and selfishness, he was analyzed by various doctors and experts to determine the cause; while unable to conclude the reasoning behind it, they speculated Stein suffered some kind of traumatic event, while the young boy only saw their attempts as inane.[48]

Chapter 21 - Young Spirit and Stein speaking about a world without god Young Stein with his weapon partner, Spirit. After joining the DWMA, he was set as Spirit Albarn's weapon partner. He would abuse his position and even beat up people not on Lord Death's list. This caused Spirit to get involved and do his best to stop him by reminding him of the rules set by Lord Death. As he adhered to Spirit's command, he claimed he wished to be born in a world without gods. However, when Spirit asked him if he was serious, he dismissed that notion, stating that such a world would be "sick".[49] Throughout his time within the academy, he would secretly experiment on Spirit's body while he was his weapon partner, which he did for five years without his knowledge.[4] Marie Mjolnir was said to have a crush on him, Stein being her "first love".[4] Eventually, his experimentation on Spirit came to be revealed by one of their classmates, whom would eventually become Spirit's next meister, wife, and eventual ex-wife. He also participated in the purging of the Star Clan. Seeing Sid Barrett thinking about taking a young baby, he questioned his decision on what to do with it. However, this earned him a chastising from his weapon partner, Spirit. He then asserted the only reason his partner felt that way was due to his own daughter being expected soon.

Perhaps Stein's most well-known weakness is his sensitivity to Madness; he himself noted that he's especially sensitive to Asura's Madness Wavelength[36] and even felt Arachne's Madness Wavelength from the Czech Republic, despite being in all the way in Death City located in Nevada.[37] This can cause him to have difficulty concentrating[38] and always puts him at risk of losing his sanity and becoming a threat to his allies. At some point his Madness becomes so profound that he is put under house arrest, and is incapacitated and unable to teach.

During his battle with Medusa Gorgon, Spirit Albarn noted that due to not having fought a serious opponent in a long time, this has caused him to "lose his edge" and also lose the ability to intimidate.[39] Despite being a powerful Meister, he's still has trouble against high-caliber opponents like Medusa. He also admitted himself while sparring with Black☆Star that the assassin knows he can get just as "jammed up" as anyone else in regular hand-to-hand combat, when he got distracted by the power of the latter's Soul Menace.
