As one of the most popular students at Monster High, Cleo enjoys a vast following, but her selection of close friends is modest and can, at its most extreme, be narrowed down to six people: Ghoulia Yelps, Deuce Gorgon, Draculaura, Frankie Stein, Lagoona Blue, and Clawdeen Wolf.

Ghoulia is one of Cleo's only confidants. The two met when Cleo reached out to her in a time of need, and they've been inseparable ever since. They make an odd couple, with Cleo being popular and Ghoulia regularly disregarded, as zombies tend to be. Even Cleo isn't always aware of her treatment of Ghoulia, treating her as a personal assistant and gateway drug, but Ghoulia knows she means well and is one of the few people Cleo will apologize to when she realizes her mistakes.

Cleo lives with her father and older sister, Nefera de Nile. Her father's name is Ramses de Nile, and he is an antiquities merchant. Cleo and Nefera's mother is Dedyet de Nile. Circumstances left her behind when the de Nile family went into hiding. The family is later reunited, according to the diaries of Hexiciah Steam and Robecca Steam, though what happens to her afterward is unknown. In The Ghoul Next Door, Nefertiti is considered the girls' aunt.

In the episode "Miss Infearmation," Cleo mentions having an older sister named Nefera de Nile, who is out of the country with their father. Clawd's "School's Out" diary also mentions Nefera as the previous cheerleading squad captain. In the "School's Out" diaries and the animated series, Cleo and Nefera don't get along very well.

Cleo de Nile has several powers inspired by Egyptian culture, such as summoning insects and speaking to snakes. Additionally:

• Glass-Shattering Scream: Cleo has been shown to be able to shatter glass with a single scream in several webisodes. • Snake Charm: Like her sister Nefera de Nile, Cleo can also charm snakes. • Enchant Scarabs: She is also supposed to be able to enchant scarabs. • Immortality: Like most undead, she is immortal. • Control of her sashes: Shown in "Welcome to Monster High", Cleo can control her sashes at will to attack and "mummify" her enemies

Cleo de Nile has black and brown hair with bangs, often enhanced with golden highlights, though she also chooses blue, teal, and/or purple as additional colors. She prefers her hair long and loose, but is not above swapping it for a shoulder-length bob. Sometimes, her hair is in a ponytail when it's long. She has tan skin and blue eyes, always surrounded by thick black eyeliner, and wears a gem on her right cheek that is blue in most of her outfits (it was red in Scaris). While Cleo can be creative with her looks, there's one fashion rule she can't break: she always has to have at least a piece of her headband on her, lest it turn to dust. Luckily for her, she can wear the headband however she sees fit. Sometimes, she wears a full bodysuit made of the material. Other times, the headbands are worked into a simple fabric at the waist, easily overlooked in the rest of the look. In Frights Camera Action, however, no headband is visible. Cleo's standard colors of choice for clothing are shades of yellow, including gold, which is her favorite color, and anything in the blue-green-teal range. She's also experimented with purple and red, but these are limited to specific periods. While not shy, Cleo prefers to wear a pair of pants, usually as part of the aforementioned jumpsuit. As part of an ancient Egyptian royal family, Cleo often incorporates large jewelry into her outfit, as well as scarabs, lotuses, snakes, and triangles (pyramids).

Cleo is a true blue-blood, so she expects everyone to treat her that way. Generally, her attitude is selfish, arrogant, sassy, ​​proud, and somewhat spoiled; she can't be entirely blamed for her behavior, as it's clear she was raised that way. She's extremely competitive with other monsters, especially Toralei Stripe. But deep down, Cleo is a very caring, kind, and considerate little monster who cares about her friends.

She can work well with others if they share a common goal; in this case, she becomes the de facto leader of the monsters. She is a highly effective organizer and leader who veers into selfish and selfless behavior depending on her mood, which is easily altered, as she can be quite temperamental when upset and violent when saddened. Furthermore, she is stubborn and will only follow her own rules if she feels like it, and she has trouble learning from her mistakes, frequently making the same mistakes of avoiding responsibility and trying to control others. It doesn't help that she has access to limited amounts of magic through her magical amulets. In the Monster High web series, Cleo is primarily known as the queen bee at school, having a self-proclaimed popularity that isn't fake, despite being self-proclaimed. Despite Cleo's general personality, one of her true personality traits is that she is kind-hearted and has a true heart of gold, which is much more than can be said for her sister Nefera, who, when asked about her best friends on her profile, simply said that she "rules alone." Cleo also uses only the name Cleo at Monster High without the addition of the surname "de Nile" to avoid raising suspicions about her name, similar to the Egyptian name Cleopatra and having a name that has to do with the river, the Nile, on which her surname is based.

Cleo was a princess of Egypt over 5,800 years ago and was raised with the knowledge that she would never become queen of Egypt, as that honor would fall to her older sister, Nefera. However, Cleo consequently had the freedom to pursue her own interests. One event that highlighted the difference in the sisters' upbringing was when a delegation from a distant country arrived and presented Nefera with a corpse flower, said to be a rare treasure, fit only for a future queen. The flower became one of Nefera's most prized possessions, while Cleo instinctively developed acrimony toward it.

Life would not be kind to the De Niles, as trouble was brewing within the kingdom. Cleo's uncle attempted to overthrow Ramses de Nile and found loyal followers among Ramses's own trusted advisors. The day the traitors staged their coup d'état had been perfectly planned to remove the royal family before anyone knew what hit them. The chief advisor took Ramses and his daughters to underground quarters, prepared in advance to shelter them in case of attack. The plan was for them to be wrapped in a magical cloth that would sustain them until the guards had dealt with the danger. Although the queen had not yet joined them, Ramses de Nile already had the family wrapped in the magical cloth that would keep them safe while his loyal followers dealt with the usurper. What they didn't know was that they no longer had any loyal followers and that the magic cloth was meant to keep them asleep forever. However, they awoke after 1,300 years, only to find the queen still missing and themselves trapped inside the hiding place. As Ramses began spending his days searching for a way out, Cleo pessimistically enjoyed the luxury of her prison, but Nefera was deeply affected by the loss of her mother and her future and spent most of her time talking to the corpse flower she had taken with her.
