Academically, Nino ranks in the bottom five of her sisters, and her strongest subject is English.

Nino has shown herself to be a good cook, from her daily meals, cookies, and conga.[10] She is able to make many friends, as noted by Yotsuba Nakano. According to Nino, the sisters have their own individual preferences, making it difficult for her to cook for them.[16]

She has poor eyesight, requiring contact lenses for her daily activities.[10] Such as when she cannot recognize Fuutarou Uesugi in a simple wig, implying her forgetful side.[3] However, this may be due to her darker surroundings and her fascination with him being blonde. The name Nino means "two" (二 ni) and "from" (乃 no). The purple rose petals on the cover of Volume 3 (Nino's volume) mean "love at first sight", which may refer to her affection for Kintarou, who is actually Futaro Uesugi. Nino owns various dolls on her bed, with one particular doll often seen in the background or as a cameo throughout the series.[17] Nino has a rabbit-themed phone case.[18] Nino dreams of opening a restaurant. Nino apparently has a fear of needles, something she shares with Itsuki Nakano. Nino prefers black tea because it can be customized to suit your taste. Nino appears to have a form of attraction towards blonde-haired people, like she was attracted to Fuutarou's younger self, and she expresses some form of attraction towards Isanari Uesugi. Ayana Taketatsu (Nino's voice actress) is the second oldest of the sisters' voice actresses.

Nino has an outgoing personality and is socially oriented, which results in her having many friends and being a social butterfly.[12] She has a sharp tongue,[10] and speaks bluntly when confronted by others, even her sisters.[10] Her rebellious attitude makes her the least agreeable girl among the sisters, making her archetype similar to the "tsundere" archetype.

Despite her cold and rude front, Nino is caring[13] and protective.[14] This is also evident in her willingness to act as the family's cook, ignoring her concerns about her own appearance (cooking activities can lead to bad odor, damaged nails, etc.). She also always carries a bandage with her, possibly as a precaution against Yotsuba Nakano's active behavior.[3]

Her interests seem to be more related to romance than anything else, such as when she wanted to watch a romance movie while Itsuki Nakano preferred a science documentary. With that in mind, she likes to openly express her feelings for Fuutarou, whether it's him or her sisters.

She also openly states that Futaro is a nobody to her and that she will deliberately do poorly in school. Also, in one of the episodes, she slapped Futaro (not just like that). She tried in every way to kick him out of her apartment, but the other sisters were against it.

According to Fuutarou, each Nakano Five has their own characteristics regarding their sheets,[15] but in Nino's case, she will carefully file her papers. Cold, sharp-tongued.

Nino is a pretty and cool-looking girl with straight, waist-length or hip-length hair (but later cut to shoulder-length), with square bangs that hang down to her eyebrows. She has reddish-pink hair, ranging from light pink to telemagnet pink[4] in Volume 1 and from antique pink to red-violet in Volume 4.[5]

Her notable accessories are two butterfly-shaped ribbons[6] on either side of her head. Like her sisters, she has dark blue eyes, is of average height, has a well-built figure, and is large-breasted. She wears a long-sleeved sweater that covers her uniform and a pair of white thigh-length socks. Both she and Ichika Nakano carry a purse.[7][8]

She is considered the most fashionable girl among the sisters, as she has various cute accessories[9] and takes care of her nails.[10] She has yet to get her ears pierced, but plans to do so at least before she gets married.[11]
