Tomoka is portrayed as shy, but she is gentle and kind. She can be very emotional, for example, not wanting to lose Donnie.

Tomoka is a sea turtle with pale green skin and white scales. Most of her hair is black, and she wears glasses. Her clothing consists of pants, a cloth around her chest, and sandals. Her hero form includes a white dress with a lilac lining, a purple bow, and her hair tied in a ponytail. Her enhanced form features a looser dress and she wears armor. Most of her long hair turns pure white. Later, her hair becomes black and white due to the effects of Aira's control over her.

Tomoka is the middle sister of the family, born to the Aeons. When they realize the Krang are going to capture her, they bring her to Earth along with her sisters. Now, as sisters and a team, her adoptive family, the O'Niles, were captured by the Krang, but she was rescued by the Turtles. Learning about the mutagen and the scientists who were captured by the Krang, they head to the facility to rescue April. She soon reunites with Shira, Manako, and Reika, and they soon live with the Turtles after they rescue Kirby.

Several months later, she and Donatello go alone to the Krang's hideout and rescue Kirby and another hostage named Anastasia, a pigeon who can communicate with the Krang.

When they accidentally released mutagen across the entire new facility after attempting to stop the Krang, Donnie and Tomoko created a retro-mutagen to return Mr. O'Neil to his normal state.
Donnie took responsibility for getting everyone to the hideout when the Krang invasion began, while Tomoko and Casey started searching for medical supplies for Shirahara's injured eye. After making their way to the farm, she and Donnie began making medicine to cure Leonardo and Shiro.

During their space adventure, Tomoka discovers she is a Princess of the Aeons in Xaava Dale. She is presented with a gift from the star Sol, a crystal, along with April's. When she connects with her crystal, her Aeon spirit, Aira senses a dark aura emanating from April, and needs her to kill Za Naron to cleanse the Earth.
